"Not only did telomerase reactivation bypass the cancer progression block that arises with telomere dysfunction, it also conferred a new property -- bone metastasis -- that was not seen in tumors that did not go through telomere dysfunction followed by telomerase reactivation," said first author Zhihu Ding, Ph.D., formerly of Dana-Farber and now with Sanofi-Aventis, Inc.
"This provides the first genetic evidence that telomerase reactivation and genome stabilization are necessary to drive full malignant progression in epithelial cancers," Ding said.
Aligning genetic alterations in mice and humans
Chin, Ding and colleagues analyzed gene copy number aberrations -- genes deleted or amplified -- in 18 advanced tumors from the mice and 194 human prostate tumors.
They found 22 of the 94 copy number alterations involving deletion or amplification of 741 genes identified in mice were similar to those found in humans. A series of analyses of changes found in bone metastases pointed to deletions of the Smad4 tumor-suppressor gene, which regulates the transforming growth factor beta (TGF-?) pathway.
The team took this finding back to the mouse model with tumor suppressors p53 and pten knocked out. These mice don't develop bone metastasis, but when the researchers also knocked out Smad4, more aggressive tumors developed, including bone metastasis in three of 24 mice.
Genes prognostic for human prostate cancer
Next, they looked at 14 genes in nine molecular pathways found to be enriched in bone metastasis to see if they were prognostic for recurrence (as measured by PSA levels after surgery) among 140 prostate cancer patients.
The 14-gene set was significantly prognostic of biochemical recurrence, providing evidence of their biological relevance to human prostate cancer, but the researchers noted that their individual contributions and mechanisms of action will require further research.
"Overall, our findings validate the integrative approach of employing genotype-phenotype correlations found in the mouse model with the power of genomic and bioinformatic analyses to discover and explain molecular mechanisms that drive prostate cancer," said co-first author Chang-Jiun Wu, Ph.D., a postdoctoral fellow in MD Anderson's Department of Genomic Medicine.

In 1960, the world population was three billion. Last year, it reached seven billion, and in 2040, the United Nations expects it to be around nine billion.
The organization noted in a recent report that the globe is running out of time to ensure that there?s enough water, energy and food for the rising population, as each year we?ll have more mouths to feed.
And the U.N. warns that if the world is unsuccessful in tackling these problems, it could send three billion people into poverty.
The Seeds for a Commodity Supercycle
According to U.N. estimates, by 2030 the globe will need at least 50% more food. And in a rapidly changing environment, new limits to supply will be created, and demand for food will rise exponentially, creating what experts call a commodity supercycle.
The current commodity supercycle, which is pushing food prices even higher, started back in 2000.
Commodity experts, like Jim Rogers, have expressed a belief that the current commodity supercycle will last for 20 to 25 years.
Chris Watling of Longview Economics agrees. Watling is the one who identified that commodity bull rallies last 20 to 25 years, tracing secular bull markets back to 1750.
And if Rodgers and Watling are correct, then the commodity bull market that started in 2000 will run all the way until 2020 to 2025.

Compared to other nitrogen fertilizers, urea is safer to transport, less corrosive to equipment and has the lowest transport cost per unit of nitrogen nutrient.
And as the major fertilizer traded internationally, urea has captured more than 56% of world trade in the dry fertilizer category to date.
Prices for urea have gradually increased over time, and are up over 360% since the beginning of 2000, costing around $368.38 per metric ton today. And prices will continue to rise as demand increases.
A New Business Model
And the small startup that could profit from increasing demand for urea fertilizers is BioNitrogen Corp. (OTC: BION.PK).
Based in Doral, Florida, BioNitrogen is a company that can produce urea fertilizer using a method that?s environmentally responsible and helpful to local economies.
More than 90% of the world?s urea production uses natural gas as the hydrocarbon feedstock. As we know, using natural gas creates carbon dioxide as a byproduct, which has a negative effect on the environment.
This is where BioNitrogen has separated itself from the competition.

The load fell into an area where crews were working, according to NBCNewYork.com. Some of the falling material?smashed a construction vehicle truck but no one was inside, The Wall Street Journal reported. One person was checked out by medical workers at the scene and released,?a New York Fire Department official told the Journal.
The beams, which?were being hoisted to the upper floors of a building project at the site, fell after a crane cable snapped, the Christian Post reported.
The size of the load and the distance it fell were not immediately known.
The accident occurred at the site of the construction of Tower 4, owned by developer Larry Silverstein, the New York Post reported. Construction of that building and at the 9/11 memorial and museum was shut down while authorities investigated, the Post reported.
?It just sounded like a massive thud. Almost like thunder,? David Stein, who was walking to his office in the Financial District when the accident happened, told the Journal. ?I heard construction workers saying that the building shook.?
Police and emergency personnel responded within minutes.
Officials are checking the stability of the site's foundation, the Fire Department said.
